The development of HIV protease inhibitors (PIs) and their inclusion in highly active antiretroviral therapies (HAARTs) marked the beginning of a treatment breakthrough in the management of human immunodeficiency virus (HIV)/acquired immunodeficiency syndrome (AIDS). The HAART treatment regimen can cut HIV viral load to undetectable levels. Nonetheless, the rapid emergence of HIV drug resistance has continued to seriously compromise long-term treatment options for HIV-infected patients. Our structure-based design strategy to develop PIs that specifically target the enzyme's backbone atoms has resulted in a number of very potent inhibitors with superior drug resistance profiles. Of particular note, our development of stereochemically defined bis(tetrahydrofuranyl) urethane as a high-affinity P2 ligand has led to the development of exceedingly potent inhibitors. One of these inhibitors, darunavir, has shown exceptional potency against the HIV-1 virus and superior activity against multi-PI-resistant viral strains. In the past decades, drug metabolizing enzymes (DME) and their role in drug interaction have been intensively investigated. Apart from DME, transporters are also involved in the absorption, distribution, and excretion of drugs; this implies that (pharmacokinetic) drug interactions may be mediated by transporters as well. However a less systematic approach is used for transporter-related drug elimination and interactions. This can be attributed to the fact that preclinical model systems to study transporter-based drug interactions differ from those typically used for metabolism- based interactions. In addition specific inhibitors and substrates are often lacking for transporters. In this research, we focused on characterizing a fluorescent transporter substrate (fluorescent bile salt analogue) with respect to its in vitro hepatobiliary disposition in various in vitro models. In addition, the utility of this probe substrate for assessment of hepatic transporter mediated drug interactions by using HIV Protease inhibitor as model interacting compounds was also investigated.

HIV-1 protease, a homodimer, has attracted the interest of many researchers due to its essential role in HIV replication and subsequent functional activities. It hydrolyses different viral proteins into their functional form to help in maturing the virus for further extending the disease propagation. The present workflow in this research was designed to identify potential HIV-1 protease inhibitors from library of approved drugs (1,428 compounds) using computational techniques in Computer-Aided-Drug-Design (CADD). The inhibitory potency of the dataset was evaluated using the lowest theoretical binding energies of the target-ligand complex. Software and tools such as Molecular Operating Environment (MOE), AutoDock Vina, Discovery Studio used in CADD were employed during the process of this work. This study suggest the possibility of repurposing some current drugs (from the library) to having potential HIV Protease inhibitory effect.

HIV-1 Proteases (PR) are an enzyme which cleaves the long HIV polyprotein into small functional peptide fragment. These small fragments again terminate into mature HIV. Inhibition of cleavage of poly protein into small peptide fragment could prevent regeneration of HIV. Computational docking between HIV-1 PR and derivatives of Cyclic Urea Inhibitors (CUIs) has provide us clue to design drug against AIDS. Computational approaches were used to dock ligand (CUIs) with HIV-1 PR to stop cleavage. CUIs are seven-member ring structure of cyclic urea compound. It incorporates the hydrogen-bonding equivalents of an enzyme-bound water molecule into a low molecular weight. Our result suggest that some of derivatives of CUIs binds with HIV-1 PR, which can be stop the fuction of HIV-1 PR(cleavage of long protein chain into small peptide fragment).Evaluation of docking results was performed based on steric complementarity. This can be called the best fitting of ligand to the enzyme pocket. The following three factors are primarily involved to influence binding conformation between ligand and a protein.These are binding energy,hydrogen bonding,and hydrophobic interaction.
